Application of Novel VEGF-Loaded PCL-P Based Hybrid Copolymer Scaffold for Bone Regeneration

Background: Polymeric materials are widely known as versatile materials, particularly for biomedical applications. The bioavailability, low toxicity, solubility, and permeability of this material play an essential role in implants, such joint replacements, bone plates, ligaments, vascular grafts, intraocular lenses, heart valves, sutures, dental implants. These can be formed either biodegradable or other naturally bioactive polymers, which composed nano-sized particles the composition materials. For example, PCL-P (HEMA-NIPAAm) is a thermosensitive, biodegradable, biocompatible hydrogel In study, we aimed to create VEGF-loaded hybrid copolymer analyze VEGF gene expression human pulp stem cells (hDPSCs) that adhere copolymer. Methods: (0.1 Â g/mL) was loaded onto synthesized (HEMA-NIPAAm), hDPSCs were seeded into scaffolds. To describe hDPSC changes, mRNA, protein, calcific deposition examined. Results: increased mRNA protein compared with any scaffold (P>0.05). After 21 days culture, color intensity alizarin red significantly higher than those (P<0.05) (P<0.05). Conclusion: Finally, thermosensitive injectable (HEMA-NIPAAM), including VEGF, enhanced genes associated osteogenesis angiogenesis hDPSCs. Thus, these results imply thermo-sensitive may good option future use carrier implantation.
